The wheel is mounted on a truing stand and adjusted for true, roundness, and tension. This should be performed regularly to keep spokes from accidentally braking on a ride (especially for downhill trail riders this is important). It should be part of your regular tune-up, but you definitely know the bike needs it if the wheel doesn't track in a straight line. The wobble will be visible.
